LAN in Borderlands 2 (and a few other games) is a bit messed up. Normally a hosted game would listen for connections on all available interfaces unless told otherwise, but not this one. It is programmed to find the adapter your system has deemed 'most important' (lowest metric value), and only use that one, doesn't matter if it's disconnected even. So if that's not the interface you're connected to each other with, it won't work without some adjustments.

Windows assigns a metric value based on the speed of the connection, which is why you normally can't play BL2 LAN over a WiFi connection. The WiFi automatically has a higher value than the wired connection. It's kind of a dummy proof thing from Microsoft's viewpoint. If you connect your computer through WiFi, but also plug in the wired connection to the same network, the lower metric value of the faster wired connection will make your computer ignore the WiFi; yay, idiot proof!
